Research Background and Purpose
During red bean paste production, large amounts of processed water are generated as the beans are repeatedly boiled.
Although this processed water is generally discarded, it may contain bioactive compounds such as polyphenols and flavonoids that are released from the beans during processing.
To evaluate its potential as a functional ingredient, this study compared the total polyphenol content, total flavonoid content, antioxidant activity, and physiological activity of the 1st, 2nd, and 3rd processed waters.

Equipment Application
The POP UV-Vis spectrophotometer was used to measure absorbance after color-development reactions.
For total polyphenol analysis, the Folin-Ciocalteu method was employed with absorbance measured at 725 nm using gallic acid as the calibration standard.
For total flavonoid analysis, the aluminum nitrate colorimetric method was used with absorbance measured at 415 nm using quercetin as the calibration standard.
These absorbance measurements enabled quantitative determination of antioxidant-related compounds in the processed water samples.

Assay Overview
The Folin-Ciocalteu reagent reacts with polyphenol compounds to form a blue-colored complex, and absorbance measured at 725 nm is used to calculate total polyphenol content (mg GAE/g).
For flavonoid analysis, flavonoids react with aluminum nitrate to produce a colored complex. Absorbance measured at 415 nm is converted into total flavonoid content (mg QE/g) using a quercetin standard curve.
These methods demonstrate how UV-Vis absorbance measurements are applied for quantitative analysis of antioxidant-related compounds.
